The present study investigates the effects of sildenafil citrate on the in vitro and in vivo activities of sildenafil citrate, a key component in sildenafil.
Sildenafil citrate has been extensively studied for the treatment of erectile dysfunction and pulmonary hypertension, and its potential is still a subject of extensive research and development. Sildenafil citrate is a phosphodiesterase-5 inhibitor with a high affinity for the cyclic GMP-specific sites of sildenafil and a high affinity for the inorganic cation-organic cation exchange site of sildenafil. The in vitro and in vivo studies indicate that sildenafil citrate inhibits phosphodiesterase type-5 (PDE5) activity and competitively inhibits PDE6. The in vivo studies indicate that sildenafil citrate has a beneficial effect on erectile function and on the vascular function, as well as on platelet aggregation. This study investigates the in vivo effects of sildenafil citrate on the function of nitric oxide (NO) and cyclic guanosine monophosphate (cGMP), which are involved in the vasodilation and vasoconstriction. Sildenafil citrate induces a decrease in the release of NO and cGMP, which are the principal enzymes responsible for degradation of cyclic guanosine monophosphate (cGMP), leading to the relaxation of smooth muscles and the reduction of blood flow to the penis. This effect is mediated by the inhibition of PDE6 activity. The results indicate that sildenafil citrate, in addition to increasing the release of NO and cGMP, induces a decrease in blood flow and increase of blood pressure in penile tissue. However, this decrease in blood pressure is not sufficient to achieve an erection when sexual stimulation is given by intracavernosal injection, because the vasodilation of penile tissue is not mediated through the activation of NO, and the blood pressure reduction occurs in response to vasoconstriction.
Sildenafil citrate has an affinity for sildenafil citrate and is not only in vitro but also in vivo. This is because sildenafil citrate has no effect on the in vitro activity of PDE-specific phosphodiesterases, and the inhibition of PDE6 has no effect on PDE-specific phosphodiesterases.
The in vitro activity of sildenafil citrate was investigated in three different doses of sildenafil citrate (3.25, 5.25 and 25.25 mg), while the in vivo activity was measured by the administration of sildenafil citrate in adult male Sprague-Dawley rats. The results indicate that sildenafil citrate has no effect on the in vitro activity of PDE-specific phosphodiesterases, and that the in vivo activity is not influenced by the dose.
